Instructions for SOLiD sequencing submissions

PLEASE NOTE: Due to continuing concerns posed by potential bioterrorism agents (select agents), our online submission form requires that you provide us with information on the original biological source of any DNA from which you will be asking us to obtain sequence. "Original biological source" refers to the organism from which the DNA was first found in nature (human, HIV, Staphylococcus aureus, etc.). If you simply put a nonspecific term such as "E. coli clone", this will not be sufficient. We realize that adding yet another field to fill out on these forms can be tiresome, but this information is necessary to protect our staff against potential legal difficulties.  Instructions for SOLiD Sequencing Submissions (01-22-08)

  1. You must submit at least 10µg of DNA for fragment libraries and 60µg of DNA for Mate-Pair Libraries.

  2. The DNA must be RNase treated and cleaned prior to submission.

  3. Include A260/280 ratio for each sample in addition to concentration total that is provided in each tube.

  4. Run the DNA you are submitting on an agarose gel and submit a copy of the picture along with your submission.You can either leave a paper copy when you drop off the sample, or you can email the picture to solid_support@ouhsc.edu. The DNA must be of high quality with no RNA contamination and little to no degradation. Samples submitted without a gel image or those showing significant degradation will not be processed.

  5. A volume of no greater than 200 µl per tube should be submitted in either TE, [pH 8.0] or nuclease free water.

  6. A current SUR, PO, or Account# must be given on the submission form. SUR's are good from the 15th of one month to the 15th of the following month and can be dropped off in BRC Rm1106 with your sample. Submissions will not be processed without a current SUR, PO, or Account#.

  7. Samples can be dropped off Monday - Friday during regular business hours (9:00 a.m. - 5:00 p.m.) in BRC Rm1106 and placed in either the brown 4ºC or the white freezer (-20 ºC).
